Background

MAP3K1 (MEKK1) is a 196-kDa serine/threonine kinase that sits at the hub of multiple signaling cascades: it functions as the apical MAP3K in the classical ERK5 and JNK pathways, phosphorylating and activating MKK5 and MKK4/7 respectively, while also cross-talking to the canonical NF-κB axis by catalyzing IKKβ phosphorylation; beyond kinase activity it acts as a ubiquitin E3 ligase that targets downstream adaptors such as TRAF2 and DUSP1 for degradation, thereby fine-tuning stress, cytokine and receptor-tyrosine-kinase responses; gain-of-function missense mutations clustered in its N-terminal PH domain and C-terminal kinase domain are among the most frequent genomic events in estrogen-receptor-positive breast and endometrial cancers, where they enhance transcription of ER-dependent proliferative genes, drive anti-apoptotic signaling and confer resistance to endocrine therapies, making MAP3K1 both a key integrator of cellular stress decisions and an actionable driver of luminal-type women’s cancers.
