Background

MAP2 (microtubule-associated protein 2) is a ~280 kDa heat-stable phosphoprotein expressed almost exclusively in neurons, where it binds along the sides of microtubules in dendrites, stabilizing them and determining dendritic caliber and branching by spacing microtubules apart; its single tandemly repeated 31-amino-acid microtubule-binding domain is regulated by phosphorylation, and proteolytic cleavage of the projection domain during excitotoxic or ischemic insults produces 40–50 kDa fragments that rapidly depolymerize dendritic microtubules, contributing to neuronal death, while antibodies to these fragments serve as widely used selective markers of dendritic cytoarchitecture in neurohistochemistry.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded mouse cerebral cortex. Anti-MAP2 antibody was used at 1/250 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Negative control: IHC shows negative staining in paraffin-embedded mouse kidney. Anti-MAP2 antibody was used at 1/250 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. Counterstained with DAPI (Blue). Heat mediated antigen retrieval with EDTA buffer pH9.0 was performed before commencing with IF staining protocol.
