Background

MANF (mesencephalic astrocyte-derived neurotrophic factor, also known as ARMET) is an evolutionarily highly conserved secreted protein, named after its initial isolation from rat midbrain astrocyte-conditioned medium and its discovery as a factor that promotes the survival of dopaminergic neurons; however, both its structure and mode of action differ from those of classical neurotrophic factors. Human MANF consists of 179 amino acids and contains an N-terminal saposin-like domain and a C-terminal SAP domain. Under normal conditions it resides mainly in the endoplasmic reticulum (ER) lumen, where it is retained through binding to BiP/HSPA5; when ER stress or hypoxia occurs, MANF expression is upregulated and the protein is secreted extracellularly. Mechanistically, MANF is a key negative regulator of the unfolded protein response (UPR): it can directly bind the ER transmembrane sensor IRE1α, reducing its oligomerization and suppressing downstream signaling such as XBP1 splicing, thereby protecting neurons from ER stress-induced death—an interaction essential for its neuroprotective effects in animal models of Parkinson's disease. Beyond the central nervous system, MANF exerts protective effects on pancreatic β cells, cardiomyocytes, retinal photoreceptors, and cells of various tissues including the liver and kidney, and MANF knockout mice develop a severe diabetic phenotype due to progressive β-cell loss. Recent studies have further revealed that after ischemic stroke, MANF expression shifts from neurons to activated microglia/macrophages, and recombinant MANF delivered intranasally or intravenously can reduce infarct volume and modulate inflammatory responses, suggesting its therapeutic potential as a broad-spectrum stress-protective and immunomodulatory factor.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human brain. Anti-MANF antibody was used at 1/800 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
