Background
Lysine malonylation (Kmal) is a dynamic and evolutionarily conserved post-translational modification in which a malonyl group, derived from the donor substrate malonyl-CoA, is covalently attached to the ε-amino group of a lysine residue. This modification dramatically alters the protein's physicochemical properties by shifting the net charge of the lysine residue from positive (+1) to negative (−1) at physiological pH, which can induce significant conformational changes and disrupt electrostatic interactions. Primarily regulated by the demalonylase SIRT5 and the acetyltransferase KAT2A, Kmal plays a crucial role in fine-tuning cellular metabolism, immune responses, and cancer progression by modifying key enzymes involved in glycolysis, fatty acid oxidation, and mitochondrial function.
