Background
LPCAT1 (Lysophosphatidylcholine Acyltransferase 1) is a membrane-bound enzyme primarily localized to the endoplasmic reticulum and Golgi apparatus, which catalyzes the acyltransferase reaction between lysophosphatidylcholine (LPC) and acyl-CoA, incorporating unsaturated fatty acids (such as arachidonic acid) into the sn-2 position of phosphatidylcholine (PC). It plays a critical role in the synthesis of pulmonary surfactant (whose major component is dipalmitoylphosphatidylcholine) and is an essential enzyme for maintaining normal lung compliance and respiratory function. However, in recent years, accumulating evidence has shown that the functions of LPCAT1 extend far beyond this, as it also plays important roles in metabolic disorders and tumors—for example, by remodeling the phospholipid composition of cell membranes, it affects membrane fluidity and signal transduction, thereby promoting tumor cell proliferation and migration. Its upregulation has been significantly associated with poor prognosis in tumors such as hepatocellular carcinoma and colorectal cancer, positioning it as an emerging anticancer drug target. Furthermore, in lipid metabolic diseases, LPCAT1 is also involved in regulating inflammatory responses and oxidative stress, playing potential roles in conditions such as atherosclerosis by influencing the generation of oxidized phospholipids.
