Background

Lipin 1 is a multifunctional phosphoprotein encoded by the LPIN1 gene, which plays a central regulatory role in mammalian lipid metabolism. As a critical "switch" in the adipogenesis process, it functions on one hand as a transcriptional coactivator in the nucleus, upregulating the expression of genes involved in fatty acid oxidation and mitochondrial biogenesis through interactions with transcription factors such as PPARα and PGC-1α; on the other hand, when localized to the endoplasmic reticulum membrane, it possesses phosphatidate phosphatase (PAP) activity, converting phosphatidic acid into diacylglycerol—a key rate-limiting step in the biosynthesis of triglycerides and phospholipids. Therefore, through this dual mechanism of nucleocytoplasmic shuttling, Lipin 1 tightly couples transcriptional regulation with enzymatic metabolic reactions, playing important roles in physiological processes such as adipocyte differentiation, hepatic lipid homeostasis, and skeletal muscle insulin sensitivity. This protein is predominantly highly expressed in skeletal muscle and adipose tissue, and its gene mutations have been confirmed to be associated with human inherited generalized lipodystrophy and severe hypertriglyceridemia, while its dysfunction is also closely linked to the development and progression of common metabolic diseases such as obesity, non-alcoholic fatty liver disease, and type 2 diabetes mellitus.
