Background

CD85j, also known as LILRB1 or ILT2, is a 110 kDa transmembrane glycoprotein belonging to the leukocyte immunoglobulin-like receptor (LILR) family. It is an inhibitory receptor broadly expressed on leukocytes, including B cells, monocytes, subsets of NK cells, memory/effector CD8+ T cells, gamma δ T cells, and monocyte-derived dendritic cells. CD85j recognizes a wide range of classical HLA-class I allelic variants, as well as non-classical molecules such as HLA-F and HLA-G, by binding to the conserved α3 domain. Additionally, it binds to the human cytomegalovirus-encoded MHC class I homologue UL18. The engagement of CD85j by its ligands induces inhibitory signals that downregulate immune responses, including the inhibition of antigen-induced proliferation and activation of CD8+ T cells, CD4+ T cells, NK cells, and gamma δ T cells. On dendritic cells, ligation of CD85j inhibits the production of cytokines like IL-10, IL-12p70, and TGF-beta and protects from Fas-mediated apoptosis.
