Background

Lhx2, also known as LIM-homeobox 2 or LH2, is a critical transcription factor belonging to the LIM-homeodomain family that plays indispensable roles in embryonic development and tissue maintenance, particularly within the central nervous system, eye, and liver. Characterized by two N-terminal LIM domains that mediate protein-pro interactions and a C-terminal homeodomain responsible for DNA binding, Lhx2 functions primarily as a nuclear regulator of gene expression, often acting in concert with cofactors such as Ldb1 to modulate chromatin accessibility and transcriptional activity. In the developing brain, it is essential for the specification of cortical identity, hippocampal formation, and the differentiation of olfactory sensory neurons, while in the eye, it governs optic cup morphogenesis and retinal progenitor cell proliferation; furthermore, recent studies have highlighted its significance in adult neurogenesis and its potential involvement in pathological conditions, including its tumor-suppressive or oncogenic duality depending on the cellular context, thereby establishing Lhx2 as a multifaceted regulator crucial for both organogenesis and cellular homeostasis.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded mouse E14 embryo. Anti-Lhx2/LH2 antibody was used at 1/4000 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
