Background

KLRG1 (Killer Cell Lectin-Like Receptor G1) is an inhibitory immune checkpoint receptor belonging to the C-type lectin-like receptor family. It is encoded by the KLRG1 gene located on human chromosome 12 and is also known by the aliases MAFA, 2F1, MAFA-L, and CLEC15A. The human KLRG1 protein consists of 195 amino acids and is a type II transmembrane glycoprotein with a molecular weight of approximately 30 kDa. Structurally, KLRG1 contains an extracellular C-type lectin-like domain (CTLD) for ligand recognition and an intracellular immunoreceptor tyrosine-based inhibitory motif (ITIM) for signal transduction.

KLRG1 is predominantly expressed on NK cells and subsets of effector and memory T cells. Its classical ligands are members of the classical cadherin family, including E-cadherin, N-cadherin, and R-cadherin. KLRG1 recognizes and binds to a highly conserved Val3-Ile7 sequence within the N-terminal EC1 domain of these cadherins via its CTLD.
