Background

KLK5 also known as stratum corneum tryptic enzyme (SCTE), is a secreted serine protease encoded by the KLK5 gene. This protein is initially synthesized as a zymogen containing a signal peptide and an activation peptide, and after proteolytic processing, it forms a mature enzyme composed of 227 amino acid residues, exhibiting typical trypsin-like activity with a preference for cleavage after arginine (Arg) residues. Under physiological conditions, KLK5 plays a central role in skin desquamation by degrading cell adhesion molecules in the stratum corneum to promote the shedding of dead cells; simultaneously, it can activate other kallikreins such as KLK7 and KLK14, forming a proteolytic cascade. In terms of pathology, dysregulation of KLK5 function is associated with various diseases: in skin disorders, hyperactivation of KLK5 is a key driver of Netherton syndrome (a severe hereditary skin disease), leading to skin barrier defects and severe inflammation; in the field of oncology, KLK5 shows aberrant expression in various cancers including ovarian cancer and breast cancer, though its exact mechanism of action remains under investigation; additionally, recent studies have found that KLK5 can be upregulated upon beta-coronavirus infection and directly cleave the viral spike protein to promote viral replication, making it a promising drug target against coronaviruses.

Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
