Background
KLHL21 (Kelch-like family member 21) is a substrate-specific adaptor protein for the Cullin 3-based E3 ubiquitin ligase complex, playing a pivotal role in regulating cellular processes through targeted protein ubiquitination and subsequent degradation. Characterized by its distinctive domain architecture, which includes an N-terminal BTB/POZ domain responsible for binding to Cullin 3 and a C-terminal kelch repeat domain that mediates specific substrate recognition, KLHL21 functions as a critical bridge between the ubiquitination machinery and its target proteins. Immunohistochemistry
IHC shows positive staining in paraffin-embedded human colon. Anti-KLHL21 antibody was used at 1/500 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
