Background
KIRREL2 (Kin of IRRE-like 2), also known as NEPH2, is a type I transmembrane protein belonging to the immunoglobulin superfamily that plays a critical role in cell-cell adhesion and signal transduction, particularly within the nervous system and kidney podocytes. Structurally characterized by extracellular immunoglobulin-like domains, a single transmembrane region, and an intracellular tail containing PDZ-binding motifs, KIRREL2 mediates homophilic and heterophilic interactions with other KIRREL family members and scaffolding proteins like CASK and ZO-1 to regulate synaptic formation, neuronal migration, and the maintenance of the glomerular filtration barrier. While it shares significant sequence homology with KIRREL1 (NEPH1) and KIRREL3, KIRREL2 exhibits distinct spatiotemporal expression patterns during development and has been implicated in neurodevelopmental disorders and renal pathologies when dysregulated, highlighting its essential function in organizing specialized intercellular junctions and translating adhesive cues into intracellular signaling cascades necessary for tissue morphogenesis and homeostasis.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded human kidney. Anti-KIRREL2 antibody was used at 1/1000 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
