Background
KIF5B and KIF5C are two major members of the Kinesin-1 family of motor proteins, encoded by distinct genes. They share a similar core structure, each containing a conserved N-terminal motor domain (which binds to microtubules and hydrolyzes ATP to generate motive force), a neck linker region, a coiled-coil stalk region (mediating dimerization), and a C-terminal tail domain (responsible for binding to various cargoes and regulation). However, they exhibit key differences in tissue distribution and function. KIF5B is widely expressed in nearly all cell types and is responsible for fundamental "housekeeping" transport processes, such as the movement of organelles like mitochondria, endoplasmic reticulum, and endosomes along microtubules. In contrast, KIF5C is predominantly and highly expressed in neurons, making it a neuron-specific kinesin critical for neuronal development and function. It is essential for the anterograde transport of specific cargoes, including presynaptic components, vesicles, and mRNAs, from the neuronal cell body along the axon to distal synapses. This transport is indispensable for maintaining neuronal polarity and synaptic plasticity. The differences in their tail domains determine their ability to recognize and bind distinct cargo adaptor proteins, enabling fine-tuned functional specialization in intracellular transport.

Flow cytometric analysis of 4% PFA fixed 90% methanol permeabilized HeLa (human cervix adenocarcinoma epithelial cell) labelling KIF5B+KIF5C antibody at 1/50 dilution (1 μg)/ (red) compared with a Rabbit monoclonal IgG (black) isotype control and an unlabelled control (cells without incubation with primary antibody and secondary antibody) (blue). Goat Anti - Rabbit IgG Alexa Fluor® 488 was used as the secondary antibody.

Immunocytochemistry
ICC shows positive staining in HeLa cells. Anti-KIF5B+KIF5C antibody was used at 1/100 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
