Background
KDM2A (also known as FBXL11 or JHDM1A) is a multifunctional protein that primarily functions as a histone H3 lysine 36 (H3K36) demethylase through its JmjC domain, specifically removing H3K36me1/2 marks. As a key epigenetic regulator, it binds specifically to unmethylated CpG islands in the genome via its CXXC zinc finger domain, thereby influencing chromatin structure and gene expression. Additionally, KDM2A contains an F-box domain that enables it to act as a substrate recognition subunit of the SCF (SKP1-Cullin-F-box) E3 ubiquitin ligase complex, participating in protein ubiquitination and degradation. Through its enzymatic activity and protein interactions, KDM2A is widely involved in regulating critical biological processes such as cell proliferation, differentiation, DNA damage repair, and maintenance of genomic stability. It also plays complex, and sometimes contradictory, regulatory roles in various cancers, including non-small cell lung cancer and breast cancer.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded mouse brain. Anti-KDM2A antibody was used at 1/100 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
