Background

KCC2 (potassium-chloride cotransporter 2) is a neuron-specific chloride-extruding transporter encoded by the SLC12A5 gene that maintains low intracellular Cl⁻ by coupling K⁺ and Cl⁻ efflux, thereby setting the hyperpolarizing, inhibitory polarity of GABA_A and glycine receptor currents essential for central nervous system inhibition, synapse maturation, dendritic spine formation, and neuronal survival, while its transcriptional and post-translational regulation (phosphorylation, membrane trafficking, activity-dependent expression) drives the developmental GABA “switch” from depolarizing to hyperpolarizing, and its down-regulation or dysfunction—triggered by epilepsy, trauma, inflammation, or genetic mutations—produces chloride accumulation, excitatory GABA, network hyperexcitability, and contributes to epilepsies, chronic pain, autism, and neurodegeneration, making KCC2 a genetically validated therapeutic target whose potentiation can restore inhibition and rescue drug-resistant neurological disorders.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human cerebral cortex. Anti-KCC2 antibody was used at 1/1000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
