Background

Jak1 (Janus kinase 1) is a 130-kDa, membrane-proximal tyrosine kinase that associates with the cytoplasmic tails of cytokine receptors—such as those for interferons, interleukins and gp130 family ligands—and, upon ligand-driven receptor clustering, trans-autophosphorylates its activation loop and then phosphorylates STAT proteins to trigger rapid gene-expression programs governing immunity, hematopoiesis and growth; because its ATP-binding cleft can be occupied by ruxolitinib, upadacitinib or other clinical inhibitors, Jak1 is a validated drug target in autoimmune diseases, myeloproliferative neoplasms and alloimmune disorders.

Immunohistochemistry

Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
