Background
ITGB7 (Integrin β7 subunit) is a critical leukocyte-specific β subunit of the integrin family, which plays a central role in lymphocyte trafficking, homing, and retention by pairing with different α subunits (particularly α4 and αE) to form heterodimers. Among these, the α4β7 integrin specifically recognizes MAdCAM-1 (mucosal addressin cell adhesion molecule-1) expressed on intestinal mucosal vascular endothelial cells, serving as the key molecular basis for mediating the homing of naïve T cells and B cells to gut-associated lymphoid tissues (such as Peyer's patches) and the intestinal lamina propria. Meanwhile, the αEβ7 integrin binds to E-cadherin as its ligand, primarily mediating the adhesion of intraepithelial lymphocytes (IELs) to intestinal epithelial cells and participating in mucosal immune surveillance. Due to the decisive role of ITGB7 in regulating intestinal immune cell trafficking, its functional pathway has become a prominent therapeutic target for inflammatory bowel diseases (such as Crohn's disease and ulcerative colitis), with antibody drugs targeting α4β7 (e.g., vedolizumab) having been successfully approved for clinical use to block aberrant lymphocyte infiltration into the intestine and thereby alleviate intestinal inflammation. Furthermore, aberrant expression of ITGB7 has also been associated with metastasis and drug resistance in certain hematological malignancies (such as mantle cell lymphoma), and its potential as both a therapeutic target and a prognostic marker is being actively explored.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded human spleen. Anti-ITGB7 antibody was used at 1/500 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
