Background

ITGA4, also known as CD49d or the α4 subunit of VLA-4, is a critical integrin α chain encoded by the ITGA4 gene. As a type I transmembrane protein, it must form a heterodimeric receptor by associating with either the β1 or β7 subunit to function on the cell surface. This integrin is an important adhesion molecule on the surface of leukocytes, capable of recognizing vascular cell adhesion molecule-1 (VCAM-1) and fibronectin, mediating firm leukocyte adhesion and transendothelial migration, thereby playing a central role in inflammatory responses and immune surveillance. Owing to its critical function in immune cell trafficking and inflammatory processes, ITGA4 has become an important clinical target for treating autoimmune and inflammatory diseases such as multiple sclerosis, Crohn's disease, and ulcerative colitis. Furthermore, studies have revealed its aberrant expression in various tumors, where it participates in tumor cell proliferation and migration by influencing signaling pathways such as PI3K-Akt, and is closely associated with patient prognosis and immunotherapeutic responses, demonstrating its potential as both a cancer biomarker and a therapeutic target.
