Background
ISG15 (Interferon-Stimulated Gene 15) is a ubiquitin-like protein that is strongly induced by type I interferons and plays a critical role in innate immunity through a post-translational modification process known as ISGylation, which involves the covalent conjugation of ISG15 to target proteins via an enzymatic cascade comprising E1 (UBE1L), E2 (UBCH8), and E3 ligases such as HERC5. Functionally, ISG15 acts as a broad-spectrum antiviral effector by modifying viral or host proteins to inhibit viral replication, modulate signaling pathways like NF-κB and IRF3, and regulate immune cell activity, while also serving extracellularly as a cytokine-like molecule that stimulates IFN-γ production from T cells and NK cells upon binding to receptors such as LFA-1; conversely, its dysregulation is implicated in various pathologies including cancer, neurodegeneration, and autoinflammatory diseases, making it both a key biomarker for interferon signatures and a potential therapeutic target for modulating immune responses.

IHC shows positive staining in paraffin-embedded human spleen. Anti-ISG15 antibody was used at 1/500 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
