Background

Osteopontin (OPN) is a multifunctional, highly acidic, calcium-binding phosphoprotein expressed ubiquitously in tissues and body fluids, where it acts as a matricellular cytokine and signaling molecule that orchestrates diverse biological processes including bone remodeling through its role as a key regulator of osteoclast function, immune responses by promoting Th1 differentiation and macrophage activation, wound healing via cell adhesion and migration, and pathological conditions such as tumor metastasis, chronic inflammation, fibrosis, and cardiovascular calcification by interacting with integrin receptors (αvβ1, αvβ3, αvβ5, αvβ6, α8β1) and CD44 variants through its conserved integrin-binding RGD motif and cryptic domains, thereby activating downstream signaling cascades like PI3K/Akt, MAPK/ERK, and NF-κB pathways that modulate gene expression for cell survival, proliferation, and tissue repair.
