Background

MHC Class I (H-2Db) is a classical class I molecule within the mouse H-2 complex, encoded by the *H2-D* locus on chromosome 17, and plays a central role in antigen presentation in the mouse immune system. Its molecular structure consists of a highly polymorphic heavy chain (α chain, containing three extracellular domains α1, α2, and α3) non-covalently associated with a β2-microglobulin light chain, with the α1/α2 domains together forming the antigen-binding groove that specifically binds and presents endogenous antigenic peptides of 9–10 amino acids in length (characterized by a canonical octamer motif, with preferential binding to peptides bearing aromatic or hydrophobic residues). Functionally, H-2Db presents intracellular viral or tumor antigens on the cell surface for specific recognition by the T-cell receptors (TCRs) of CD8⁺ cytotoxic T lymphocytes (CTLs), thereby triggering CTL-mediated killing effects—a key mechanism for the clearance of infected and tumor cells. Notably, H-2Db holds significant tool value in research: it is one of the most commonly used MHC molecules for tetramer staining techniques, leveraging its high-affinity binding to specific antigenic peptides (such as GP33-41 from LCMV or SIINFEKL from ovalbumin) to enable precise tracking and quantification of antigen-specific CD8⁺ T-cell responses. Furthermore, alongside H-2Kb and H-2Lb, H-2Db is a major classical class I molecule, but allelic differences across mouse strains directly determine the strength and specificity of T-cell responses, making it an indispensable key molecule in transplantation rejection, autoimmune diseases, and tumor immunology research.
