Background
ICOSL (CD275), also known as B7-H2, is a ~60 kDa transmembrane glycoprotein of the B7/immunoglobulin superfamily that delivers the key costimulatory signal to ICOS on activated T cells, thereby driving T-cell proliferation, Th1/Th2/Th17 cytokine secretion, germinal-center formation, plasma-cell differentiation and memory T-cell survival, while also undergoing reverse signaling into dendritic cells, B cells, endothelial cells or tumor cells to modulate adhesion, migration and cytokine output, and—uniquely in humans—additionally engaging CD28 and CTLA-4 to fine-tune both naïve and regulatory T-cell responses, positioning it as a central checkpoint in adaptive immunity, allergy, autoimmunity and cancer.
