Flow cytometric analysis of Mouse CD19 expression on C57BL/6 mouse splenocytes. Cells were labeled with either Rat IgG2a, κ Isotype Control (black line histogram) or SDT Rat Anti-Mouse CD19 antibody (red line histogram) at 1/500 dilution (1 μg). Goat Anti-Rat IgG Alexa Fluor® 488 was used as the secondary antibody. Total viable cells, as determined by Fixable Viability Dye 545 (S0B88802), were used for analysis. Flow cytometry and data analysis were performed using Agilent NovoCyte Quanteon and FlowJo™ software.

Background
CD19 is a 95 kDa type I transmembrane glycoprotein that belongs to the immunoglobulin superfamily and is expressed on the surface of B lymphocytes and follicular dendritic cells. It plays a crucial role in B cell activation and humoral immune responses by forming a complex with CD21, CD81, and CD225. This complex reduces the threshold for B cell receptor (BCR) activation, enhancing downstream signaling pathways and facilitating B cell responses to antigens. CD19 is also a biomarker for B cell development and lymphoma diagnosis, and it is targeted in immunotherapies for diseases like leukemia.
