Background
Intelectin-1 (also known as omentin-1 or ITLN1) is a soluble lectin primarily secreted by intestinal goblet cells and adipose tissue, existing as a circulating protein in human plasma. Its structure contains a characteristic fibrinogen-related domain and multiple glycosylation sites. It specifically recognizes and binds to microbial glycan antigens containing furanose residues (such as galactofuranose), exerting innate immune defense functions through bacterial agglutination or direct pathogen clearance. Additionally, Intelectin-1 participates in adipose tissue metabolism by inhibiting adipogenesis and regulating adiponectin secretion, thereby maintaining glucose homeostasis and insulin sensitivity. In clinical studies, its expression levels are closely associated with various diseases: in obesity and type 2 diabetes mellitus, the expression of Intelectin-1 in adipose tissue is downregulated, and it is considered a protective factor against metabolic disorders. In inflammatory bowel diseases such as ulcerative colitis, intestinal Intelectin-1 expression is elevated, participating in mucosal barrier repair and anti-inflammatory responses. Furthermore, Intelectin-1, as a major component of eosinophil extracellular trap networks, plays a pathological regulatory role in asthma and allergic diseases, and its genetic variants are significantly associated with susceptibility to non-alcoholic fatty liver disease. Notably, Intelectin-1 is also a potential biomarker for certain tumors; in colorectal cancer, its upregulated expression is negatively correlated with tumor differentiation. In summary, Intelectin-1 possesses triple functions of immune defense, metabolic regulation, and inflammatory modulation, serving as a key molecule connecting the intestinal barrier, adipose tissue, and systemic metabolic homeostasis.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded human colon. Anti-Intelectin-1 antibody was used at 1/1000 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Anti- Intelectin-1 antibody was used at 1/1000 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin.
