Background

INPP4B (inositol polyphosphate 4-phosphatase type II) is a lipid phosphatase that selectively dephosphorylates PI(3,4)P₂ to PI(3)P, thereby acting as a brake on the PI3K/AKT signaling cascade by limiting AKT plasma-membrane recruitment and activation; through this mechanism it functions context-dependently as either a tumor suppressor—restraining proliferation, survival and chemoresistance in breast, prostate, gastric and nasopharyngeal cancers—or, paradoxically, as an oncogenic facilitator via SGK3 activation in subsets of melanoma, colon cancer and AML, while in male germ cells it safeguards DNA integrity and mitochondrial function during spermiogenesis.
