Background

SMARCB1, also known as INI1 or BAF47, is a core component of the human SWI/SNF chromatin remodeling complex, which regulates gene transcriptional activity by altering chromatin structure. As a 47-kilodalton protein composed of 385 amino acids and localized in the nucleus, SMARCB1 can bind to DNA and plays critical roles in various life processes, including cell proliferation, differentiation, and tumor suppression. The protein is best known for its function as a tumor suppressor, and mutations or loss of its expression are closely associated with the development of multiple aggressive tumors. For instance, biallelic inactivation of SMARCB1 is a key molecular event leading to rhabdoid tumors—highly malignant neoplasms that predominantly affect infants and young children—and its germline mutations also underlie the rhabdoid tumor predisposition syndrome (RTPS1). Furthermore, variations in the SMARCB1 gene have also been linked to schwannomatosis (a benign nerve sheath tumor disorder) and Coffin-Siris syndrome (a hereditary developmental disorder), demonstrating the diversity of disease spectrums resulting from its functional abnormalities.
