Background

Inhibin alpha is a 366-residue glycoprotein that dimerizes to form the common α-subunit of the inhibin family; it is synthesized primarily in Sertoli cells of the testis and granulosa cells of the ovary, where it partners with either a βA or βB chain to create inhibin A or inhibin B, both of which act as potent negative-feedback regulators of pituitary FSH secretion by antagonizing activin signaling through competition for the type II activin receptor, thereby governing gametogenesis and steroidogenesis; beyond reproduction, inhibin alpha is a sensitive serum tumor marker for granulosa-cell and Sertoli-cell neoplasms, and its expression is controlled by gonadotropins, local growth factors, and epigenetic mechanisms, making it a key node in the hypothalamic–pituitary–gonadal axis.

Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Heat mediated antigen retrieval with EDTA buffer pH9.0 was performed before commencing with IF staining protocol.

Negative control: IF shows negative staining in paraffin-embedded human kidney. Anti-Inhibin alpha chain antibody was used at 1/200 dilution and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. Counterstained with DAPI (Blue). Heat mediated antigen retrieval with EDTA buffer pH9.0 was performed before commencing with IF staining protocol.
