Background
Interleukin-31 (IL-31) is a cytokine that belongs to the IL-6 family of inflammatory cytokines. It is primarily produced by type 2 helper T-cells (TH2) and plays a significant role in triggering cell-mediated immunity against pathogens. IL-31 signals through a receptor complex composed of IL-31 receptor A (IL-31RA) and oncostatin M receptor β (OSMRβ), which are expressed in immune and epithelial cells. These receptors activate multiple signaling pathways, including ERK1/2 MAP kinase, PI3K/AKT, and JAK1/2, thereby influencing immune responses and cell survival.
In mice, IL-31 is particularly notable for its involvement in chronic inflammatory diseases, such as atopic dermatitis. Studies have shown that IL-31 expression is significantly upregulated in mouse models of atopic dermatitis, similar to its role in human skin lesions. Additionally, IL-31 has been implicated in pruritus (itching) through its effects on sensory neurons, particularly those expressing TRPV1 and TRPA1.
