Background
Interleukin-31 (IL-31) is a novel cytokine belonging to the IL-6 family, primarily produced by activated T helper 2 (Th2) cells and mast cells. It signals through a heterodimeric receptor composed of IL-31 receptor A (IL-31RA) and oncostatin M receptor β (OSMRβ), which is expressed on various cell types including sensory neurons, keratinocytes, and immune cells. IL-31 is a key mediator of pruritus (itch) in inflammatory skin diseases such as atopic dermatitis, psoriasis, and allergic conditions. Its signaling pathway promotes itch sensation, disrupts epidermal barrier function, and drives immune responses, making it a significant therapeutic target for chronic pruritic disorders.
