Background

Interleukin-23 (IL-23) is a heterodimeric cytokine composed of p19 and p40 subunits. Its functional receptor, IL-23R, combines with the IL-12Rβ1 subunit to form a type I cytokine receptor complex. This complex activates the downstream JAK2-STAT3 signaling pathway, regulating the proliferation, differentiation, and secretion of inflammatory factors in immune cells such as T cells and NK cells. It plays a central role in sustaining Th17 cell-mediated inflammatory responses. Clinically, abnormal activation of the IL-23/IL-23R pathway is a key pathogenic mechanism in autoimmune diseases such as psoriasis and inflammatory bowel disease. Additionally, it promotes tumorigenesis within the tumor microenvironment, making it a critical therapeutic target.
