Background
IL-2 receptor gamma chain (IL-2 Rγ, CD132) is the 64 kDa shared signaling subunit of the hematopoietin receptor family that converts low-affinity IL-2 binding into a high-affinity, signal-competent complex. Independently, CD132 does not bind IL-2, yet its cytoplasmic tail is obligatory for recruitment of Janus kinase 3 (JAK3) and subsequent activation of STAT5, PI3K and MAPK cascades that drive T-, B- and NK-cell survival, proliferation and differentiation. The same γ-chain is also exploited by IL-4, IL-7, IL-9, IL-15 and IL-21 receptors, explaining why germ-line mutations in IL2RG produce X-linked severe combined immunodeficiency.
