Background
IL-2Rβ (CD122) serves as the common β subunit shared by both the interleukin-2 receptor (IL-2R) and the IL-15 receptor, forming a dimeric intermediate-affinity receptor with IL-2Rγ (CD132) or a trimeric high-affinity receptor with CD25 (IL-2Rα) and CD132, thereby mediating IL-2 and IL-15 signal transduction through the JAK-STAT pathway. It plays a central role in regulating the development, proliferation, survival, and differentiation of lymphocytes such as T cells and NK cells, particularly influencing CD8⁺ T memory cell maintenance, invariant natural killer T (iNKT) cell generation, and thymocyte development. Clinically, CD122 gene mutations can lead to severe immune dysregulation and peripheral tolerance defects, while aberrant expression is closely associated with immune dysfunction in ANCA-associated vasculitis, systemic lupus erythematosus, inflammatory bowel disease, and HBV/HCV infections. In recent years, CD122-biased IL-2 agonists (e.g., NKTR-214/bempegaldesleukin, ANV419) have emerged as a hotspot in cancer immunotherapy, selectively binding the CD122/CD132 dimer to preferentially activate CD8⁺ T cells and NK cells while reducing regulatory T (Treg) cell-mediated immune suppression, thus offering novel precision therapeutic strategies for cancer and autoimmune diseases.
