Background
Interleukin-19 (IL-19) is an immunosuppressive cytokine belonging to the IL-10 subfamily, encoded by the IL-19 gene located on chromosome 1. It is preferentially expressed in monocytes, macrophages, and T and B lymphocytes. IL-19 binds to the IL-20 receptor complex, activating the JAK-STAT signaling pathway, which promotes the Th2 (regulatory) T-cell response, supports an anti-inflammatory lymphocyte phenotype, and inhibits the production of pro-inflammatory cytokines like IFN-γ. It also suppresses the expression of RNA-binding protein HuR, reducing the translation of cell adhesion molecules and limiting vascular tissue damage. However, IL-19 has also been reported to enhance chronic inflammatory diseases, as it regulates cells of the monocyte lineage and affects processes like cell differentiation.
