Background

IL-18BP (Interleukin-18 Binding Protein) is a secreted immunoregulatory glycoprotein belonging to the IL-1 receptor family, whose structure contains a single Ig-like domain that binds IL-18 with high affinity (Kd≈400 pM) and blocks its interaction with the IL-18Rα/β receptors, thereby inhibiting NF-κB and AP-1 signaling pathways and downstream IFN-γ production; recombinant IL-18BP His-tagged protein derived from Cynomolgus monkeys holds critical clinical significance in drug development, serving as a natural antagonist for treating NLRC4-related autoinflammatory diseases, rheumatoid arthritis, and other Th1-mediated autoimmune disorders, while its identification as a secreted immune checkpoint has revealed the resistance mechanism of IL-18 immunotherapy, driving the development of next-generation biologics including anti-IL-18BP antibodies and IL-18 mutants, with this protein also serving as a biomarker for radiation injury and a potential target for radioprotective agents.

Bioactivity

Measured by its ability to inhibit the IL-18-induced response of KG-1 human acute myelogenous leukemia cells. The IC50 for this effect is 12-21  ng/mL in the presence of 20 ng/mL of recombinant human IL-18.
