Background
IL-17RB (Interleukin-17 Receptor B), also known as IL-25R, is a type I transmembrane glycoprotein belonging to the IL-17 receptor family. It serves as the specific receptor subunit for the cytokine IL-25 (also called IL-17E) and forms a functional signaling complex with IL-17RA.
Mechanism of Action: IL-25 binds specifically to IL-17RB, which allosterically initiates the formation of a ternary complex with IL-17RA, necessary for downstream signaling events . This complex recruits the adaptor protein ACT1 and activates canonical NF-κB and MAPK pathways, leading to the production of type 2 cytokines such as IL-4, IL-5, and IL-13 . The IL-25/IL-17RB axis is a critical driver of type 2 immune responses.
