Background
IL-17RA and IL-17RC serve as the principal receptor subunits for IL-17 family cytokines, forming a classic heterodimeric signaling complex. Structurally, both receptors contain fibronectin type III domains in their extracellular regions and engage IL-17A/F ligands via a tip-to-tip assembly mode to form ternary complexes, which further organize into 2:2:2 hexameric signalosomes to amplify downstream signaling. Functionally, this receptor complex drives pro-inflammatory responses through the activation of NF-κB and MAPK pathways, playing a pivotal role in the pathogenesis of autoimmune diseases such as psoriasis and ankylosing spondylitis. Clinically, brodalumab (anti-IL-17RA) and anti-IL-17A monoclonal antibodies targeting this axis have been widely applied in the treatment of inflammatory disorders, while small-molecule inhibitors are currently under active development.
