Background
IL-17RA and IL-17RC serve as the principalreceptor subunits for IL-17 family cytokines, forming a classic heterodimericsignaling complex. Structurally, both receptors contain fibronectin type IIIdomains in their extracellular regions and engage IL-17A/F ligands via atip-to-tip assembly mode to form ternary complexes, which further organize into2:2:2 hexameric signalosomes to amplify downstream signaling. Functionally,this receptor complex drives pro-inflammatory responses through the activationof NF-κB and MAPK pathways, playing a pivotal role inthe pathogenesis of autoimmune diseases such as psoriasis and ankylosingspondylitis. Clinically, brodalumab (anti-IL-17RA) and anti-IL-17A monoclonalantibodies targeting this axis have been widely applied in the treatment ofinflammatory disorders, while small-molecule inhibitors are currently under activedevelopment.
