Background

IGSF3 (Immunoglobulin Superfamily Member 3), also known as EWI-3 or V8, is a type I single-pass transmembrane protein encoded by the IGSF3 gene. Its structure comprises an N-terminal signal peptide and eight immunoglobulin-like domains, and it is primarily localized to the cell membrane with abundant expression in tissues such as kidney, placenta, and lung. Although its structure is highly similar to CD101, which is involved in T cell activation, IGSF3 is not expressed in lymphocytes, and its physiological functions are currently not fully understood, though it is speculated to function as a cell surface receptor or cell adhesion molecule. Its functional dysregulation is closely associated with specific diseases: homozygous mutations in the IGSF3 gene have been confirmed to cause autosomal recessive bilateral nasolacrimal duct obstruction (congenital lacrimal duct defect); meanwhile, recent studies have found that IGSF3 is significantly overexpressed in hepatocellular carcinoma tissues and can promote tumor cell proliferation, migration, and invasion through activation of the NF-κB signaling pathway, suggesting its potential as a prognostic biomarker or therapeutic target for liver cancer.
