Background

Insulin-like Growth Factor Binding Protein-4 (IGFBP-4) is the smallest member of the IGFBP family, with a molecular weight of approximately 24 kDa, encoded by the IGFBP4 gene located on human chromosome 17q12-21.1. The protein consists of 237 amino acids and contains the conserved N-terminal, C-terminal, and variable L-domains characteristic of the IGFBP family, including two additional interconnected cysteine residues in the L-domain. IGFBP-4 binds both IGF-I and IGF-II with high affinity, exhibiting 2–10 fold higher affinity for IGF-II than for IGF-I. This binding prolongs IGF half-life and inhibits ligand-receptor interactions, thereby attenuating IGF-mediated proliferation and differentiation in most physiological contexts.

Bioactivity

Measured by its ability to inhibit the biological activity of IGFII on MCF7 human breast adenocarcinoma cells. The EC50 for this effect is typically 31-48ng/mL in the presence of 14 ng/mL human IGFII.
