Background
Insulin-like Growth Factor Binding Protein-4 (IGFBP-4) is the smallest member of the IGFBP family, with a molecular weight of approximately 24 kDa, encoded by the IGFBP4 gene located on human chromosome 17q12-21.1. The protein consists of 237 amino acids and contains the conserved N-terminal, C-terminal, and variable L-domains characteristic of the IGFBP family, including two additional interconnected cysteine residues in the L-domain. IGFBP-4 binds both IGF-I and IGF-II with high affinity, exhibiting 2–10 fold higher affinity for IGF-II than for IGF-I. This binding prolongs IGF half-life and inhibits ligand-receptor interactions, thereby attenuating IGF-mediated proliferation and differentiation in most physiological contexts.
