Background

Insulin-like growth factor-binding protein-2 (IGFBP-2) is a 36-kDa cysteine-rich secreted glycoprotein that preferentially binds IGF-II and is highly expressed during embryogenesis, especially in the central nervous system and reproductive tissues. Beyond simply sequestering IGFs, IGFBP-2 possesses intrinsic, IGF-independent activities: it contains an RGD integrin-binding motif, is internalized via a nuclear-localization signal, and can activate PI3K/Akt, NF-κB and β-catenin pathways. Consequently, IGFBP-2 functions as a developmental regulator and, when aberrantly re-activated, as an oncogenic hub that promotes epithelial-to-mesenchymal transition, angiogenesis, stemness and chemo-resistance in multiple solid tumours.
