Background
Hyaluronidase PH20, also known as SPAM1 (Sperm Adhesion Molecule 1), is a multifunctional glycosyl hydrolase enzyme belonging to the glycosyl hydrolase 56 family. It is a GPI-anchored protein expressed in the testis and found on the surface of mammalian sperm and the inner acrosomal membrane. PH20/SPAM1 is unique for its hyaluronidase activity at neutral pH, which is essential for sperm to penetrate the hyaluronic acid-rich cumulus cell layer surrounding the oocyte during fertilization. Additionally, it plays roles in zona pellucida binding and calcium signaling associated with acrosomal exocytosis. This enzyme is also involved in sperm maturation and has potential applications in reproductive medicine and contraception.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded human testis. Anti-Hyaluronidase PH20/SPAM1 antibody was used at 1/1000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
