Background

HuR/ELAVL1 protein, also known as human antigen R, is a ubiquitously expressed RNA-binding protein encoded by the ELAVL1 gene. It is a member of the ELAV family of proteins and contains three RNA recognition motifs (RRMs) that enable it to bind to specific RNA sequences, particularly AU-rich elements (AREs) in the 3' untranslated regions (3'UTRs) of mRNAs. This binding stabilizes the mRNA, protecting it from degradation and enhancing its translation, thereby regulating gene expression. HuR plays crucial roles in various cellular processes, including cell differentiation, stress response, and tumorigenesis. Its dysregulation has been implicated in multiple diseases, such as cancer, cardiovascular diseases, and neurodegenerative disorders.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human cerebral cortex. Anti-HuR/ELAVL1 antibody was used at 1/1000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunocytochemistry

ICC shows positive staining in HEK293 cells. Anti-HuR/ELAVL1 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
