Background
Human WIF1 (Wnt inhibitory factor 1) is a secreted glycoprotein that functions as a key antagonist of the canonical Wnt signaling pathway by directly binding to Wnt ligands. It exhibits the highest expression in the lung and plays critical roles in embryonic development, particularly in lung morphogenesis. WIF1 acts as a tumor suppressor, and its expression is frequently silenced by promoter hypermethylation in various human cancers, leading to aberrant Wnt activation. Elevated plasma WIF‑1 levels have also been associated with pulmonary hypertension in heart failure patients, suggesting its potential as a prognostic biomarker. Restoring WIF1 expression represents a promising therapeutic strategy for cancers where it is epigenetically inactivated.
