Background
Vascular endothelial growth factor (VEGF) is a subfamily of growth factors that play a critical role in stimulating angiogenesis—the formation of new blood vessels. As a key signaling protein, VEGF is involved in both embryonic vascular development and the physiological or pathological growth of existing vasculature. The VEGF family comprises several members, including VEGF-A, VEGF-B, VEGF-C, VEGF-D, and placental growth factor (PlGF), with VEGF-A being the first identified. Prior to the discovery of other isoforms, VEGF-A was commonly referred to as VEGF. Due to its central role in promoting tumor angiogenesis, VEGF is essential for tumor development and metastasis, making it a pivotal target in cancer therapeutics. Accumulating evidence indicates that single nucleotide polymorphisms (SNPs) within the VEGF gene may serve as predictive and prognostic biomarkers for major solid tumors, such as breast cancer, non-small cell lung cancer, colorectal cancer, and prostate cancer.

Linearity
The concentrations of Human VEGFA were measured and interpolated from the target standard curves and corrected for sample dilution.
#1 sample is undiluted samples are as follows: human plasma-Heparin (100%). The interpolated dilution factor corrected values are plotted. The mean target concentration was determined to be 141.18 pg/mL in human plasma-Heparin.
#2 sample is undiluted samples are as follows: human plasma-Citrate (100%). The interpolated dilution factor corrected values are plotted. The mean target concentration was determined to be 243.03 pg/mL in human plasma-Citrate.
#3 sample is undiluted samples are as follows: A549 supernatant (100%). The interpolated dilution factor corrected values are plotted. The mean target concentration was determined to be 162.55 pg/mL in A549 supernatant.
The sample is undiluted samples are as follows: A431 supernatant (50%). The interpolated dilution factor corrected values are plotted. The mean target concentration was determined to be 5888.62 pg/mL in A431 supernatant.
To assess the linearity of the assay, two samples were spiked with high concentrations of Human VEGFA in various matrices and diluted with the appropriate Calibrator Diluent to produce samples with values within the dynamic range of the assay.
