Background
Human triggering receptor expressed on myeloid cells 2 (TREM2) is a transmembrane glycoprotein belonging to the immunoglobulin superfamily that is selectively expressed on microglia in the central nervous system, as well as on macrophages, dendritic cells, and osteoclasts in the periphery. The full-length human TREM2 protein consists of 230 amino acids, comprising an extracellular immunoglobulin-like domain, a short stalk region, a transmembrane domain, and a cytoplasmic tail. Functionally, TREM2 regulates microglial survival, activation, phagocytosis, and inflammatory responses, playing a critical role in maintaining brain homeostasis. Its ectodomain undergoes proteolytic shedding by metalloproteases such as ADAM10 and ADAM17, releasing a soluble form (sTREM2) that can be detected in cerebrospinal fluid and blood. TREM2 is one of the strongest genetic risk factors for late-onset Alzheimer's disease, with loss-of-function variants such as R47H significantly increasing disease risk. The receptor engages multiple ligands, including apolipoprotein E, amyloid-β, and charged lipids, through distinct binding surfaces on its extracellular domain. Recombinant human TREM2 is widely used in research to study microglial biology, neuroinflammation, and Alzheimer's disease pathogenesis.

ELISA
Spike Recovery
The recovery of Human TREM2 was evaluated in activated samples spiked with concentrations spanning the entire assay range.
Spike Recovery
Recovery of Human TREM2 was assessed by spiking a 500 pg/mL standard into 3% human serum. Recovery of Human TREM2 was assessed by spiking a 500 pg/mL standard into 5% human EDTA plasma. Serial dilutions of the spiked sample were prepared, and the percent recovery was calculated for each dilution relative to the expected concentration.
Cross-reactivity
The heatmap displays OD450 ratios for serum samples from multiple species. Color intensity is proportional to the O.D.450 ratio (test species / reference species). Darker colors indicate higher cross-reactivity.
Spike-and-dilution Linearity
To evaluate assay linearity, three samples were spiked with high levels of Human TREM2 in cell culture media and serially diluted with the corresponding Calibrator Diluent to fall within the assay's dynamic range.
Determination of Minimum Detectable Dose (MDD)
The MDD was determined using three independent lots of assay components. For each lot, 19 replicate measurements of the diluent (zero calibrator) were performed. The mean (AVERAGE) and standard deviation (STDEV) of the 19 replicates were calculated. The MDD for each lot was then calculated according to the following formula:
MDD = 2 × STDEV + AVERAGE
HOOK Effect Threshold
The upper limit of the HOOK effect was established at 100× the highest calibrator concentration (equivalent to a 2-log10 increase). No HOOK effect was observed below this threshold, confirming that samples within this concentration range yield reliable quantitative results without signal depression.
Intra-Assay Precision (Precision within an assay)
Three samples of known concentration were tested fifteen times on one plate to assess intra-assay precision.
Inter-Assay Precision (Precision between assays)
Three samples of known concentration were tested in separate assays to assess inter-assay precision. Assays were performed with at least three lots of components.
Dilution Linearity
The concentrations of Human TREM2 were measured and interpolated from the target standard curves and corrected for sample dilution.
The leftmost bar in the graph represents neat biological samples. "Undiluted" was defined as the neat biological sample (100%). The subsequent bars represent two-fold serial dilutions of this reference. The mean target concentration was determined to be 191.1 pg/mL in the unstimulated THP-1 cell supernatant.
Dilution Linearity
The concentrations of Human TREM2 were measured and interpolated from the target standard curves and corrected for sample dilution.
The leftmost bar in the graph shows biological samples diluted in Assay Diluent to 50% concentration. "undiluted" was defined as 50% biological sample. The subsequent bars represent two-fold serial dilutions of this reference. The mean target concentration was determined to be 2983.3 pg/mL in stimulated THP-1 cell supernatant.
Dilution Linearity
The concentrations of Human TREM2 were measured and interpolated from the target standard curves and corrected for sample dilution.
The leftmost bar in the graph shows biological samples diluted in Assay Diluent to 3% concentration. "undiluted" was defined as 3% biological sample. The subsequent bars represent two-fold serial dilutions of this reference. The mean target concentration was determined to be 12540 pg/mL in human serum.
Dilution Linearity
The concentrations of Human TREM2 were measured and interpolated from the target standard curves and corrected for sample dilution.
The leftmost bar in the graph shows biological samples diluted in Assay Diluent to 5% concentration. "undiluted" was defined as 5% biological sample. The subsequent bars represent two-fold serial dilutions of this reference. The mean target concentration was determined to be 11292.9 pg/mL in human EDTA plasma.
Serum levels in healthy individuals
Duplicate measurements were performed on serum samples obtained from ten individual healthy donors. Data are reported as dilution factor-corrected interpolated. The resultant mean TREM2 concentration was 5107.4 pg/mL, with a distribution spanning 0 to 9841.2 pg/mL across the donor cohort.
Comparison of Standard Curves from Two Kits
Standard curves from Company A and Starter Onestep were compared.
