Background

Human Tau (phospho T181) refers to the Human Tau protein specifically phosphorylated at the threonine residue in position 181. This particular post-translational modification is a critical epitope in the study of Alzheimer's disease and other tauopathies. In a pathological context, the hyperphosphorylation of Tau at sites like Thr181 reduces its affinity for microtubules, contributing to cytoskeletal destabilization and promoting the protein's aggregation into neurofibrillary tangles. Crucially, this phosphorylated form is not just an intracellular pathological marker; it is normally secreted into the interstitial fluid and can ultimately enter the cerebrospinal fluid and blood. As a result, Tau phosphorylated at Thr181 (p-Tau181) has gained immense clinical significance as a core biomarker in the diagnosis and monitoring of Alzheimer's disease. Elevated levels of p-Tau181 in both cerebrospinal fluid and, more recently, blood plasma have been strongly correlated with the presence of Alzheimer's-type pathology, specifically tau deposition, helping to differentiate it from other neurodegenerative conditions.
