Background
Human and monkey TNF RI (TNFRSF1A) are highly homologous type I transmembrane receptors that share a conserved extracellular cysteine-rich ligand-binding domain, a transmembrane helix, and an intracellular death domain responsible for mediating NF-κB, MAPK, and apoptotic signaling upon TNF-α stimulation. Due to their structural and functional similarity, monkey TNF RI serves as an essential translational model in non-human primate preclinical studies for evaluating drug efficacy and safety, while human TNF RI is a major therapeutic target for autoimmune and inflammatory diseases, with applications including recombinant protein production, antibody development, small-molecule inhibitor screening, and structure-based drug design aimed at modulating its signaling pathways.
