Background

Human lysozyme (LZM/LYZ), a 14.4-kDa bacteriolytic enzyme encoded by the LYZ gene on chromosome 12, functions as a critical innate immune defender by hydrolyzing β-1,4-glycosidic bonds in peptidoglycan of bacterial cell walls. Primarily secreted by neutrophils, macrophages, and epithelial cells, it is abundantly present in secretions (tears, saliva) and biological fluids, with serum/plasma concentrations typically ranging from 1.5–3.0 μg/mL in healthy adults. Clinically, elevated serum levels (>5 μg/mL) serve as a biomarker for bacterial infections (e.g., tuberculosis, meningitis), monocytic leukemia (due to malignant cell release), and renal tubular dysfunction (reduced reabsorption), while decreased levels correlate with immunosuppression. Detection relies on high-sensitivity ELISA (limit: ~0.5 ng/mL), though sample handling requires rigorous centrifugation and -80°C storage to prevent degradation. Its dual role as an antimicrobial agent and disease indicator underscores its significance in both physiological immunity and pathological diagnostics.
