Background
Human interleukin-8 (IL-8), also known as CXCL8, is a key CXC chemokine that functions as a central mediator of the inflammatory response. It is primarily produced by immune cells (such as macrophages) and tissue cells (like epithelial and endothelial cells) upon stimulation by infection or injury. IL-8’s most prominent role is as a potent chemoattractant and activator for neutrophils, guiding these white blood cells from the bloodstream to inflamed tissues by binding to its receptors, CXCR1 and CXCR2. Beyond recruiting neutrophils, IL-8 also promotes angiogenesis and enhances neutrophil antimicrobial functions. While essential for effective host defense, dysregulated or excessive IL-8 production is closely associated with the pathogenesis of numerous inflammatory diseases—including chronic obstructive pulmonary disease, rheumatoid arthritis, and inflammatory bowel disease—and contributes to tumor progression and metastasis in cancer, making it a significant biomarker and potential therapeutic target.

The concentrations of Human IL-8 were measured and interpolated from the target standard curves and corrected for sample dilution.
#1 sample is undiluted samples are as follows: human PBMC cells stimulated with 10ug/ml PHA for 5days was 4000fold dilution. The interpolated dilution factor corrected values are plotted. The mean target concentration was determined to be 120.17 ng/mL in stimulated human PBMC supernatant.
#2 sample is undiluted samples are as follows: human PBMC cells culture for 5days was 4000fold dilution. The interpolated dilution factor corrected values are plotted. The mean target concentration was determined to be 183.29 ng/mL in unstimulated human PBMC supernatant.
To assess the linearity of the assay, the sample was spiked with high concentrations of Human IL-8 in various matrices and diluted with the appropriate Calibrator Diluent to produce samples with values within the dynamic range of the assay.
