Background
IL-12 is a key immunoregulatory cytokine produced primarily by immune cells like macrophages and dendritic cells. Its fundamental role is to bridge innate and adaptive immunity. It potently stimulates the activation of natural killer (NK) cells and T cells. A major function is driving the differentiation of naive T cells into the T-helper 1 (Th1) type. This action prompts T and NK cells to produce interferon-gamma (IFN-γ), a critical cytokine for cell-mediated immunity. Consequently, IL-12 is essential for defending against intracellular pathogens and tumors. It also enhances the cytotoxic activity of immune cells, enabling them to destroy infected or malignant targets. Due to its potent effects, its activity is tightly regulated within the body. Dysregulation of IL-12 is linked to autoimmune diseases, highlighting its powerful inflammatory potential. In summary, IL-12 is a master regulator of pro-inflammatory immune responses.
